ABSTRACT

ObjectiveTo observe the effect of Danshen injection (DAN) on platelet (PLT)-induced metastasis of breast cancer cells in vitro. MethodThe 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to observe the effect of DAN on the growth of MDA-MB-231 cells in vitro. Oris™ migration assay was used to determine the effect of DAN (final mass concentrations 4, 8, 16 g·L-1) on PLT (1.5×1010 cells/L)-induced migration of breast cancer cells in vitro. The effect of DAN on PLT-induced cell invasion was detected by Transwell assay. Immunofluorescence and Western blot were used to detect the effect of DAN on the protein expression associated with PLT-induced epithelial-mesenchymal transition (EMT). In addition, enzyme-linked immune-sorbent assay (ELISA) was used to determine the effect of DAN (final mass concentrations 4, 8, 16, 32, 64 g·L-1) on the secretion of transforming growth factor-β1 (TGF-β1). Western blot was used to observe the effect of DAN on the expression of podoplanin (PDPN) protein in MDA-MB-231 cells induced by PLT. ResultCompared with the blank group, the DAN groups (32 and 64 g·L-1) showed decreased A570 (P<0.05, P<0.01), and there was no significant difference in A570 between DAN groups (4, 8, 16 g·L-1). Compared with the blank group, the PLT group showed increased cell migration and invasion, while DAN groups significantly inhibited PLT-induced cell migration and invasion. Compared with the blank group, the PLT group showed decreased expression of E-cadherin, while DAN could significantly reverse this effect of PLT. Compared with the blank group, the PLT group showed increased Slug and Snail protein expression (P<0.05, P<0.01), while DAN significantly reversed Snail protein expression induced by PLT (P<0.05, P<0.01). The content of TGF-β1 in the PLT group increased (P<0.01), while the secretion of TGF-β1 induced by PLT decreased in the DAN groups (16, 32, and 64 g·L-1) (P<0.05, P<0.01), and the secretion of TGF-β1 was not significantly affected in other DAN groups. PDPN protein expression in the PLT group increased (P<0.01), while DAN could significantly inhibit PLT-induced PDPN expression (P<0.01). ConclusionDAN can inhibit PLT-induced migration, invasion, and EMT of breast cancer cells. The mechanism may be related to the direct action between breast cancer cells and tumor cells by down-regulating PDPN expression and interfering with PLT and has nothing to do with the effect of TGF-β1 secretion of PLT.

ABSTRACT

AIM: To explore the efficacy of Danshen injection iontophoresis in the treatment of retinal vein occlusion(RVO).METHODS: The clinical data of 90 patients(90 eyes)with RVO treated in our hospital from January 2020 to December 2021 were analyzed retrospectively, and they were divided into control group and observation group according to treatment methods. A total of 38 patients(38 eyes)in the control group were treated with intravenous Xueshuantong+oral compound Xueshuantong capsule+enteric-coated aspirin, and 52 patients(52 eyes)in the observation group were treated with Danshen injection iontophoresis on this basis. The treatment time of the two groups was 3mo. The clinical efficacy, best corrected visual acuity, retinal condition and hemorheological indexes(whole blood low shear viscosity, fibrinogen)were compared between the two groups before and after treatment.RESULTS: The total effective rate of the observation group was significantly higher than that in the control group(87% vs 58%,P&#x003C;0.05)at 3mo after treatment. The best corrected visual acuity in both groups was better than that before treatment, and the observation group was better than the control group(all P&#x003C;0.05).The circulation time of retinal vein, the relative diameter of retinal vein and the relative area of retinal hemorrhage in the observation group were lower than those before treatment,and the observation group was lower than the control group(all P&#x003C;0.05). The whole blood low shear viscosity and fibrinogen levels in the two groups were lower than those before treatment, and the observation group was lower than those in the control group(all P&#x003C;0.05).CONCLUSION: Danshen injection iontophoresis is effective in the treatment of patients with RVO, which can effectively improve the visual acuity, treat fundus lesions and improve the abnormal blood flow.

ABSTRACT

Objective:To investigate the inhibitory effects of Danshen injection against ovarian cancer cell proliferation induced by the interaction between platelets and cancer cells. Method:The induction of platelets on SKOV3 growth <italic>in vitro</italic> and the inhibitory effect of Danshen injection at 12,24,and 48 g·L<sup>-1</sup> were observed by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and cell colony formation assays. The content of transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>) in the platelet-tumor cell interaction system and platelet supernatant and the effect of Danshen injection on TGF-<italic>β</italic><sub>1 </sub>secretion were detected by enzyme-linked immunosorbent assay (ELISA). The influences of tumor cell culture supernatant on platelet aggregation and secretion and the inhibitory effect of Danshen injection were determined by microplate assay and ELISA. The effects of Danshen injection on platelet nuclear factor kappa B (NF-<italic>κ</italic>B) signaling pathway were assayed by Western Blot. Result:Compared with the blank group, the platelet induction group exhibited significantly elevated absorbance at <italic>A</italic><sub>570 </sub>(<italic>P</italic><0.01), while the absorbance at <italic>A</italic><sub>570</sub> in the platelet + Danshen injection group was significantly lower than that in the platelet induction group (<italic>P</italic><0.01). The comparison with the Danshen injection group revealed that the cell proliferation inhibitory rate in the platelet + Danshen injection group at the same dose was more significant (<italic>P</italic><0.01). The number of colonies in the platelet induction group was obviously increased in contrast to that in the blank group(<italic>P</italic><0.05), while the number of colonies in the platelet + Danshen injection group was significantly lower than that in the platelet induction group(<italic>P</italic><0.05,<italic>P</italic><0.01). As demonstrated by comparison with the blank group, TGF-<italic>β</italic><sub>1 </sub>content in the supernatant of the platelet induction group rose remarkably(<italic>P</italic><0.01), whereas that in the platelet + Danshen injection group declined(<italic>P</italic><0.01). Compared with the Danshen injection (24 g·L<sup>-1</sup>) group, the platelet + Danshen injection group displayed more obvious inhibition(<italic>P</italic><0.01). Compared with the blank group, Danshen injection significantly reduced the TGF-<italic>β</italic><sub>1 </sub>content in platelet supernatant(<italic>P</italic><0.05,<italic>P</italic><0.01). There was no significant change in the content of TGF-<italic>β</italic><sub>1 </sub>in SKOV3 supernatant treated with Danshen injection. The platelet aggregation, thromboxane A<sub>2</sub>(TXB<sub>2</sub>), and serotonin (5-HT) secretion in the SKOV3 cell supernatant induction group were significantly increased as compared with those in the blank group (<italic>P</italic><0.01), while such indexes in the cell supernatant induction + Danshen injection group were obviously decreased (<italic>P</italic><0.01). Compared with the Danshen injection (24 g·L<sup>-1</sup>) group, the cell supernatant induction + Danshen injection group displayed more obvious inhibition at the same dose(<italic>P</italic><0.01). Compared with the blank group, the platelet induction group exhibited obviously up-regulated phosphorylated TGF-<italic>β</italic>-activated kinase-1 (TAK-1) and NF-<italic>κ</italic>B, but down-regulated phosphorylated inhibitory protein of NF-<italic>κ</italic>B (I<italic>κ</italic>B)(<italic>P</italic><0.01), which however were significantly reversed in the platelet + Danshen injection group<bold>(</bold><italic>P</italic><0.01). Conclusion:Danshen injection affect the proliferation of SKOV3 cells by inhibiting their interaction with platelets, which may be related to the inhibited secretion of TGF-<italic>β</italic><sub>1</sub>.

ABSTRACT

To evaluate the efficacy and safety of Compound Danshen Injection combined with Western medicine in the treatment of vascular dementia. Databases of Cochrane Library, PubMed, EMbase, CNKI, SinoMed, VIP, Wanfang Data were electronically retrieved for collecting randomized controlled trial(RCT)about vascular dementia treated with Western medicine alone or combined with Compound Danshen Injection from the year of database establishment to January 2020. Two researchers independently screened out li-teratures, extracted data, and evaluated the risk of bias for inclusion in the study. Meta-analysis was conducted using RevMan 5.3 software. A total of 5 RCTs were included, involving 588 patients, with 299 in treatment group and 289 in control group. Meta-analysis results showed that compared with Western medicine alone, Compound Danshen Injection combined with Western medicine was better in the effective rate(RR=1.23,95%CI[1.14,1.33],P<0.000 01), MMSE score(MD=3.54,95%CI[3.01,4.06],P<0.000 01), ADL score(MD=11.49,95%CI[8.05,14.93],P<0.000 01), the level of CRP(MD=-0.72,95%CI[-1.25,-0.20],P=0.007) and the level of IL-6(MD=-7.64,95%CI[-9.65,-5.63],P<0.000 01). Adverse reactions mainly included rash and skin prick, which did not affect the treatment effect. Based on the findings, the combination of Compound Danshen Injection in the treatment of vascular dementia could improve the effective rates, relieve the mental state damage and improve the daily living ability, with mild adverse reactions and a low incidence. However, due to the low quality of the included literatures, high-quality and large-scale randomized controlled trials are needed for further verification.

ABSTRACT

OBJECTIVES: This study aims to compare the clinical efficacy of an integrated approach to prevent and treat the recurrence of moderate-to-severe intrauterine adhesions (IUA) after hysteroscopic transcervical resection of adhesion (TCRA). METHODS: The study included a total of 70 patients with moderate-to-severe IUAs who underwent TCRA. Patients were randomly divided into two groups: treatment group (n=35) and control group n=35). In the treatment group, patients underwent balloon uterine stent placement and artificial cycle as well as received intrauterine perfusion of Danshen injection and oral Chinese medicine. In the control group, patients underwent balloon uterine stent placement and artificial cycle as well as received hyaluronic acid sodium and intrauterine device (IUD). Follow-up was performed after treatment of uterine cavity, menstruation and pregnancy. RESULTS: After 3 months of treatment, we observed a significantly lower rate of intrauterine re-adhesion (45.71% versus 77.14%, p=0.044) and significantly higher clinical efficiency (82.86% versus 77.14%, p=0.025) in the treatment group than those in the control group. After 6 months of treatment, we observed a significantly higher clinical efficiency in the treatment group than that in the control group (88.57% versus 68.57%, p=0.039). During the follow-up period, the pregnancy rate was 45.71% and 37.14% in the treatment group and control group, respectively, although the difference was not statistically significant (p=0.628). CONCLUSIONS: After surgical management of IUA, the integrated treatment combining a uterus stent placement and artificial cycle with Danshen injection and oral Chinese medicine can improve the condition of menstruation, and prevent and treat recurrence of IUA.

ABSTRACT

Testing and analysis of chemical markers is currently the prevailing approach for quality assessment of traditional Chinese medicines. However,several important issues remain to be addressed,including the trade-off between accuracy and coverage. In this study,in order to give full play to the advantages of their respective methods and provide technical support for more comprehensively and rapidly evaluate the quality of Danshen Injection products,a fingerprint method was coupled with quantitative analysis of multicomponents by single marker( QAMS),with Danshen Injection as the carrier. Ultra performance liquid chromatography( UPLC) was used to establish the quantitative fingerprint. The UPLC fingerprints contained 13 common peaks,11 of which were identified by using ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry( UPLC-QTOF-MS). Furthermore,with sodium danshensu as the internal reference substance,relative correction factors( RCFs) of protocatechuic aldehyde,caffeic acid,rosmarinic acid,lithospermic acid,and salvianolic acid B were calculated through slope analysis method,and the QAMS method was adopted to determine the contents of these 6 components. The UPLC fingerprint was employed to assess the consistency of 12 batches of Danshen Injection,which showed good batch-to-batch consistency with similarity higher than 0. 99. In the comparison of contents of the six constituents obtained by QAMS and external standard method( ESM),RSD was all less than 4. 3%,indicating the good accuracy of the QAMS method. The QAMS method developed in this study combined with UPLC fingerprint can comprehensively reflect the internal quality of Danshen Injection when only the reference substance sodium danshensu is consumed,with greatly reduced detection cost and time. It provides a technical basis for further improving the quality standard of Danshen Injection.

Objective·To explore the mechanism of Danshen injection on promoting spinal cord functional recovery by observing the expression of brain-derived neurotrophic factor (BDNF) and insulin-like growth factor-1 (IGF-1) in rats with spinal cord injury (SCI). Methods·Thirty SD rats were divided randomly into normal control group, model group and treatment group (n=10). The rats in model group and treatment group were subjected to weight-drop SCI according to the Allens' method, and administered normal saline and Danshen injection, at the dosage of 1 mL/kg once a day , by intraperitoneal injection respectively for 14 days. The combine behavioral score (CBS) of rats in all groups was detected on 1, 3, 7 and 14 days after SCI, the expression of BDNF and IGF-1 in all groups was detected with immunohistochemical method and Western blotting. Results·Compared with the normal control group, the CBS in model group and treatment group was elevated after SCI, and declined gradually with the lapse of injury time in different rates. Until 14 days after SCI, compared with the model group, the CBS in treatment group was lower (P=0.000), and the expression levels of BDNF and IGF-1 (including immunohistochemistry positive cell number and relative content) in treatment group were both elevated significantly (all P<0.05). Conclusion·Danshen injection can promote the recovery of nerve function by elevating the expression of BDNF and IGF-1 after SCI.

ABSTRACT

Objective To analyse effects of Compound Danshen injection on acute viral myocarditis.Methods-128 patients with acute viral myocarditis were divided into two groups from May 2015 to February 2017 in Yuhang District, the Second People's Hospital.The two groups received the conventional treatment plan, the observation group was given compound Danshen injection, the evaluation of compound Danshen injection on the disease effect.Results①Total efficiency of observation group was 93.75%,higher than that of control group 76.56%(P<0.05).②Serum TNF-ɑ and IL-6 level of observation group were[(72.4±10.4), (36.15±4.68)]ng/L,lower than that of control group[(95.0±12.7), (46.23±5.24)ng/L](P<0.05).③Serum AST,CK-MB and LDHI level of observation group were[(43.19±3.25), (17.34±3.31), (168.4±13.9)]U/L,lower than that of control group[(0.47±0.08)μg/L, (9.05±1.93)mg/L)](P<0.05).Whole blood viscosity,plasma viscosity and FIB level of observation group were[(5.95±1.91)mPa·s, (1.49±0.17)mPa·s, (3.20±0.58g/L)],lower than that of control group[(52.09±4.61), (25.55±3.86),(201.5±15.4)U/L](P<0.05).④There was no obvious adverse reaction in the control group and the observation group had 3.13% adverse reactions.ConclusionCompound Danshen injection can reduce the level of inflammation in patients with acute viral myocarditis,protect myocardial cells and improve treatment effect.

ABSTRACT

Objective To observe the effect of Danshen injection combined with ulinastatin on the brain protection and inflammatory reaction in patients with Severe traumatic brain injury.Methods 83 cases of patient with Severe traumatic brain injury were randomly divided into the observation group(43 cases)and the control group(40 cases).The control group was given ulinastatin based on conventional treatment,and the observation group was given Danshen injection and ulinastatin based on the control group.For 10 days,the differences of hemodynamic parameters(Qmean、Zc、DR)and serum nerve function indexes(β-EP、DynAl-13、NSE),inflammatory factors(IL-1,IL-6,TNF-α,CRP)between the two groups were be compared.Results ①Comparison of hemodynamic parameters There was significantly difference of Qmean、Zc、DR in these two groups(Fgroup=6.126,5.024,5.621,P<0.05),Qmean was showed a rising trend over time and Zc、DR were showed a declining trend over time(Ftime=10.146,9.247,9.381,P<0.05),the changed amplitude in observation group was higher than the control group(Finteraction=5.357,4.257,4.813,P<0.05); Comparison of nerve function indexes There was significantly difference of β-EP、DynAl-13、NSE in these two groups(Fgroup=5.827,6.294,6.731,P<0.05),they was all showed a reducing trend over time(Ftime=12.613,11.746,9.842,P<0.05),the declined amplitude in observation group was higher than the control group(Finteraction=6.353,7.251,4.284,P<0.05);Comparison of inflammatory factors There was significantly difference of IL-1,IL-6,TNF-α,CRP in these two groups(Fgroup=4.284,5.162,6.174,4.291,P<0.05),they was all showed a rising then reducing trend over time(Ftime=9.163,10.357,13.457,12.434,P<0.05),the changed amplitude in observation group was higher than the control group(Finteraction=4.255,5.242,8.530,7.847,P<0.05).Conclusion Danshen injection combined with ulinastatin has more obviously function of protecting the cerebralfunction and suppressing inflammation in the treatment of Severe traumatic brain injury compared with ulinastatin alone.

ABSTRACT

Objective:To compare the effectiveness and safety of Dengzhanhuasu and Danshen Injection in the treatment of the patients with angina pectoris of coronary heart disease with Meta-analysis, and to provide basis for their application in clinic.Methods:Computer search was performed using Cochrane Central Register of Controlled Trial,Ovid-medline,EMBASE,PubMed,CNKI,Wanfang,VIP and CBM,and the literatures about the effectiveness and safety of Dengzhanhuasu and Dansheng Injection in the treatment of the patients with angina pectonis of coronary heart disease were obtained.According to the principle of selection and elimination, the literatures were selected, and the qualities of literatures were evaluated.The related data were extracted from the literatures.RevMan 5.3 software was used to extract the data for Meta-analysis.Results:A total of 167 literatures were retrieved, and 8 randomlized-controlled studies were selected according to the selection criteria.The evaluation results of the effectiveness of angina pectoris according to symptom showed that χ2=2.62,df=7,P=0.92,Z=6.85 (P<0.000 01), 95%CI:2.73-6.10. The evaluation of the effectiveness of angina pectoris according to electrocardiogram,χ2=6.37,df=7,P=0.50,Z=6.37(P=0.000 01),95%CI:1.94-3.49.The results of Meta-analysis showed that there were significant differences in the relief of symptoms and improvement of the electrocardiogram efficacy between Dengzhanhuasu and Danshen Injection in the treatment of angina pectoris of coronary heart disease (P<0.05).Conclusion:Dengzhanhuasu has better effectiveness in the treatment of the patients with unstable angina than Dansheng Injection and they have the same safety.

ABSTRACT

This study was aimed to investigate the effect of Compound Danshen injection in treatment of dysphagia by electromyography (EMG) guided on new Renying point caused by bulbar paralysis after stroke.A total of 60 patients with dydphagia caused by cerebral palsy after stroke were randomly divided into the experimental group and the control group (30 patients in each group).Patients in the control group were conducted with routine treatment,local low frequency electrical stimulation and rehabilitation training.Patients in the experimental group were treated with acupoint injection therapy guided by EMG on the basis of treatment of the control group.Four weeks were one treatment course.Observations were made on changes of indexes,such as water swallow test,swallowing ability evaluation,and etc.between two groups.The results showed that the cure rate of the experimental group was 76.67%,which was higher than that of the control group (50.00%).The treatment efficiency of the experimental group (93.33%) was higher than that of the control group (80.00%),with statistical significance (P < 0.05).It was concluded that clinical effects of EMG guided injection on new Renying point in the treatment of dysphagia caused by bulbar paralysis in the experimental group was better than that of the control group.It is worthy widely applied and distributed in clinical practices.

ABSTRACT

Objective To investigate the efficacy of ultrasound-guided nerve block anesthesia combined with Danshen injection on the treatment of angina and its influence on serum indexes. Methods 178 patients with angina in Huzhou central hospital from July 2015 to February 2017 were randomLy divided into the control group (n=89) and the research group (n=89). The control group was given routine western medicine combined with Danshen injection, while the research group was treated with ultrasound-guided nerve block anesthesia combined with Danshen injection. The clinical efficacy, homocysteine (Hcy), adiponectin (APN), tumor necrosis factor-α (TNF-α), high-sensitivity C-reactive protein (hs-CRP), ejection fraction (EF) before and after treatment, adverse reactions were compared between the two groups. Results The total effective rate in the control group (80.90%) was lower than that in the control group (95.51%) (P<0.05); After treatment, the levels of Hcy, TNF-α, hs-CRP in the two groups increased, and the variety range in the research group was larger (P<0.05); The incidence of adverse reactions in the control group and the research group (3.37% VS 1.12%) were compared, there was no statistical difference. Conclusion Ultrasound-guided nerve block anesthesia combined with Danshen injection could effectively improve serum indexes of patients, it has definite efficacy and less adverse reactions, which is worthy of further clinical application and promotion.

Objective To investigate the efficacy of ultrasound-guided nerve block anesthesia combined with Danshen injection on the treatment of angina and its influence on serum indexes. Methods 178 patients with angina in Huzhou central hospital from July 2015 to February 2017 were randomLy divided into the control group (n=89) and the research group (n=89). The control group was given routine western medicine combined with Danshen injection, while the research group was treated with ultrasound-guided nerve block anesthesia combined with Danshen injection. The clinical efficacy, homocysteine (Hcy), adiponectin (APN), tumor necrosis factor-α (TNF-α), high-sensitivity C-reactive protein (hs-CRP), ejection fraction (EF) before and after treatment, adverse reactions were compared between the two groups. Results The total effective rate in the control group (80.90%) was lower than that in the control group (95.51%) (P<0.05); After treatment, the levels of Hcy, TNF-α, hs-CRP in the two groups increased, and the variety range in the research group was larger (P<0.05); The incidence of adverse reactions in the control group and the research group (3.37% VS 1.12%) were compared, there was no statistical difference. Conclusion Ultrasound-guided nerve block anesthesia combined with Danshen injection could effectively improve serum indexes of patients, it has definite efficacy and less adverse reactions, which is worthy of further clinical application and promotion.

ABSTRACT

<p><b>OBJECTIVE</b>To systematically evaluate the clinical effectiveness and safety of Danshen Injection (, DS) as one adjuvant treatment for conventional therapy with Western medicine (WM) for unstable angina pectoris (UAP).</p><p><b>METHODS</b>Using literature databases, a thorough and systematic retrieval of randomized controlled trials (RCTs) comparing DS plus WM with WM was conducted from inception to April 2015. The extracted data from included studies was analyzed by Review Manager 5.2 software. The Cochrane risk of bias tool was used to assess the quality of included studies, and Begg's and Egger's tests conducted by Stata 12.0 were used to evaluate the potential presence of publication bias.</p><p><b>RESULTS</b>A total of 17 RCTs, which involving 1,433 participants, were identified and reviewed. The meta-analysis indicated that the combined use of DS and WM was significantly superior to WM alone for UAP in terms of the total effectiveness rate of angina pectoris [risk ratio (RR) =1.23, 95% confidence interval (CI): 1.17, 1.29, P<0.01] and the total effectiveness rate of electrocardiogram (ECG) [RR=1.18, 95%CI: 1.06, 1.30, P=0.001]. Additionally, DS could also further reduce the content of fibrinogen, adjust blood lipid level, correct T wave inversion, and so on. Fifteen adverse drug reactions were reported in two studies, Four of which appeared in the experimental group.</p><p><b>CONCLUSION</b>Based on the systematic review, the combined use of DS and WM was more effective than WM alone, it can be further widely used in clinic, however, there was no exact conclusion for its safety.</p>

ABSTRACT

OBJECTIVE:To obverse the efficacy and safety of tolynicate and napthylacetic acid and atorvastation combined with Danshen injection in the treatment of alcoholic hyperlipemia syndrome. METHODS:21 patients with alcoholic hyperlipemia syndrome were enrolled. All patients were given maintaining acid-base balance,oxygen inhalation and other conventional treatment. Based on it,they were orally given 75 mg Tolynicate and napthylacetic acid tablet,3 times a day+20 mg Atorvastatin calcium tab-let,once a day+4 ml Compound danshen injection,adding into 150 ml 5% Glucose injection by intravenous infusion,once a day. The treatment course for both groups was 30 d. Clinical efficacy,ALT,AST,TBIL,TC,TG,RBC and Hb before and after 6, 12,18,24 and 30 d,and incidence of adverse reactions were observed. RESULTS:After treatment,the total effective rate was 95.3%and incidence of adverse reactions was 28.5%. The ALT,AST and TBIL after 6,12,18,24 and 30 h and TC and TG after 18,24 and 30 h were significantly lower than before and gradually decreased by time;RBC and Hb after 12,18,24 and 30 h were significantly higher than before and gradually increased by time,the differences were statistically significant(P<0.05). CON-CLUSIONS:tolynicate and napthylacetic acid and atorvastation combined with Danshen injection is effective in the treatment of al-coholic hyperlipemia syndrome,with good safety.

ABSTRACT

Objective: To clarify the cardioprotective effects of Yiqi Yangyin Huoxue (YYH) Injection exert against ischemia-reperfusion (I/R) injury, and the mechanisms involving activation of specific survival signals reperfusion injury salvage kinase (PI3K-Akt pathway and ERK1/2 pathway), hence inhibiting mitochondrial permeability transition pore opening (mPTP). Methods: A Langendorff model of myocardial ischemia-reperfusion injury was employed. The rats were randomly divided into seven groups: control group, I/R group, YYH 2.5, 5, and 10 μL/mL groups, LY294002 + YYH 10 μL/mL group, and LY294002 group. The cardiac parameters of left ventricular developed pressure (LVDP), maximal rise/fall of left ventricular pressure (± dp/dtmax), and rate-pressure product (RPP) were monitored. Creatine kinase (CK) and lactate dehydrogenase (LDH) released from effluents were measured. Infarct size was estimated by TTC staining. Transmission electron microscopy (TEM) was performed to assess morphological difference between cardiac mitochondrial isolated I/R rats and YYH pretreated rats. Western blotting was used to determine some protein expression. MPTP opening in mitochondria isolated from Langendorff rat hearts pretreated with YYH was measured. Incubation of isolated cardiac mitochondria with YYH and mPTP opening was measured. Results: YYH 10 μL/mL can significantly improve ventricular function, ameliorate the level of myocardial tissue lesions. YYH (5 and 10 μL/mL) reduced CK and LDH release. And these protections were accompanied by a significant increase in p-PKB/Akt, p-ERK1/2, and p-GSK-3β (Ser-9). LY294002 abolished the protective effects of YYH on cardiac function and infarct size and inhibited YYH-induced phosphorylation of PKB/Akt and GSK-3β. MPTP opening in mitochondria isolated from YYH-pretreated occurred more readily in vitro than I/R mitochondria. YYH (2.5, 5, 10, and 20 μL/mL), Shenmai injection (2.5, 5, 10, and 20 μL/mL) resulted in a significant inhibition of Ca2+-induced mitochondrial swelling. Conclusion: YYH produces direct cardioprotective actions. Inhibition of mPTP is a key event by which it mediates myocardial protection against I/R injury. And this effects involve activation of PI3K-Akt and ERK1/2 signal pathway. The mechanism of its action is different from that of CsA.

Objective To study the therapeutical effects of compound Danshen injection combined with edaravone in treatment of acute cerebral infarction.Methods 130 cases of ACI were randomly divided into two groups, control group(65 case) and observation group(65 cases), control group were administered with edaravone injection, while observation group were administered with compound Danshen injection and edaravone injection, NIHSS score and Barthel index score were used for effects evaluation, meanwhile, serous levels of copeptin, NT-proBNP and Hcy were measured by ELISA assay.Results Observation group had a better effect on ACI care, compared with control group (P<0.01), and serum levels of copeptin,NT-proBNP and Hcy significantly improved both in the two groups ( P <0.01 ) , while, observation group had a better effects ( P <0.01 ) . Conclusion Combination therapy of compound Danshen injection and edaravone has a fabulous effect on ACI care.

ABSTRACT

OBJECTIVE:To observe clinical efficacy and safety of naloxone hydrochloride combined with Danshen injection in the treatment of neonatal hypoxic ischemic encephalopathy(HIE). METHODS:104 patients with HIE were randomly divided into observation group and control group with 52 cases in each group. The control group was treated with Naloxone hydrochloride injec-tion 0.02 mg/kg,ivgtt,qd and conventional symptomatic treatment;while observation group was additionally treated with Danshen injection 4-6 ml+10% Glucose injection 20 ml,ivgtt,qd,on the basis of the control group. Treatment course of 2 groups lasted for 7 d. Clinical efficacy,symptom and sign recovery of 2 groups were compared,as well as levels of serum MMP-9,IL-6 and TNF-α,cerebral hemodynamic parameters and ADR. RESULTS:There was no statistical significance in MMP-9,IL-6,TNF-α and cerebral hemodynamic parameters before treatment(P>0.05). The total effective rate of observation group(92.31%)was signifi-cantly higher than that of control group (75.00%);after treatment,the recovery time of consciousness disturbance,primitive re-flex and muscle tension in the observation group were shorter than those in the control group;the levels of serum MMP-9,IL-6 and TNF-α in observation group were significantly lower than those in control group;peak systolic flow velocity(PSFV),end-dia-stolic flow velocity (EDTV) were higher in the observation group than in the control group,with statistical significance (P0.05). No ADR was ob-served in 2 groups. CONCLUSIONS:Naloxone hydrochloride combined with Danshen injection can significantly promote HIE and signs recovery,reduce the levels of serum MMP-9,IL-6 and TNF-α,and improve hemodynamic parameters with good safety.

Objective To study the effect of Danshen injection on brain peptide type B (NT‐proBNP) in patients with hyperten‐sion and left ventricular failure ,provide an effective basis for clinical treatment .Methods Totally 80 patients of hypertension with left heart failure from February 2015 to February 2016 were randomly divided into two groups :40 patients were treated with con‐ventional therapy (control group ) ,40 patients in the conventional treatment combined with Danshen injection (experimental group) .We compared NT‐proBNP ,inflammatory factors and clinical efficacy between two groups .Results Before treatment ,the expression of NT‐proBNP and hs‐CRP ,NT‐proBNP level was basically the same in two groups (P> 0 .05) .After treatment ,the ex‐perimental group of patients with NT‐proBNP was significantly lower than the control group and the LVEF ,left ventricular mass index and left ventricular volume index were significantly higher compared with the control group (P< 0 .05) .The total effective rate of patients in Danshen injection groups was significantly higher (P< 0 .05) .Conclusion The injection of Danshen in patients with hypertension and heart failure patients can significantly improving NT‐proBNP levels and improve the cardiac function .
